Sugar and water is insufficient- cells need other materials to make proteins and such.

The urea is there as a nitrogen source, which is necessary for amino acid biosynthesis and can be used as an energy source. I'm not sure if ammonium can be used for energy, but it's also a source of nitrogen and the phosphate is another necessary nutrient (ATP = adenosine tri phosphate).

Apple juice has sugar, phosphate and sufficient trace amounts of peptides or other nitrogen sources for cells to grow.

For a kitchen media, I think the easiest thing to do would be to use yeast extract. Alternatively (and even easier) you could buy premade media mix. Looks like it's $37.50 for 5L worth of mix.

Also- make sure you're sterilizing whatever media you make in a pressure cooker before using it. It's a poor man's autoclave.

None are found locally. Not sure how size matters in terms of quality, it's the same material. When I'm doing transfers for isolation, the extra 15mm from the center is hardly used since the transfers are made before they even get close to the edges. I bought these before the glass ones and made all my transfers long before they fully colonised.

I haven’t actually done this with spores, but I have worked with agar quite a bit so next time I get myself a syringe I’ll definitely be doing this. It shouldn’t be that hard, but you need to work in a SAB or under a Bunsen Burner or a similar sized flame. Google proper agar inoculation techniques. Here’s a link to some agar plates.

If anyone wants to start using agar, but doesn’t want to make their own, this is the agar I buy when I’m not making it myself. Knock on wood, but I have never had a contam on any of the 20 plates I’ve gone through.

Its only $20. I think you would be hard pressed to make equivalent media at home. If you are really cheap you can use agar agar from an Asian grocery and pour plates but buying the dishes, and processing the plates will cost nearly as much as just buying the premade plates.

Your doctor is not going to change your meds based on one fasting blood glucose level. It's gonna take months to get your A1C down to the normal range of 6.0 or lower (about 120mg/dl blood glucose average).

You will need agar. You can make your own, but to start you can buy agar plates on Amazon.

Here is a cloning tek that will buy you a couple of extra days if you are waiting on the agar.

Edit: Do a search for "mushroom cloning teks" and look at some others. They may be better for you.
