So! it's been shown that Yotsuba has been making cranes for a long while, early on in the chapters we see her making a bunch of well made cranes for "a friend of a friend who is in the hospital". This is despite being new to the school / recently on the basketball team, though this is beside the point. While she was making the cranes for Uesugi, she mentions that she's also making it for the "1000 paper cranes" and we learn that while cranes are common knowledge in japan (through Nino's and I think Ichika's responses), they don't know the story behind them.As Negi's a writer, he also does a lot of research to add elements to stories and I believe that this is also one of them.

The Tl;dr of the legend is that making 1000 cranes = 1 wish granted by the gods. Some stories grant happiness and eternal luck instead of just 1 wish, there's also stories where instead of just 1000 cranes in total, there's a condition where they have to be made in 1 year.

After this point I thought yeets was just going to make a wish- until I got to the story part that popularized the legend

Sadako Sasaki tldr

A japanese girl who was exposed to radiation at the age of 2, by 12 she developed leukemia, got hospitalized and began trying to make 1000 paper cranes because of the legend

From here, her friends and family have said that she completed the 1000 and continued after even when her wish didn't come true

Then there's the fictionalized storybook version where she only folded 644 before passing away. her friends and family completing them afterwards.

I was still a little skeptic here, then I looked up the storybook and read the synopsis

Instead of our Yots getting afflicted with leukemia, we get the quints Mom afflicted with acute leukemia.

We know little about the quints mother, we learn that she grows ill and gets hospitalized. Then passes away on the 14th of august the year they meet Uesugi.In raising 5 of them I believe their mom passed off her symptoms of the cancer as exhaustion until the disease progressed worse and worse until she got hospitalized and at the same time, meeting PapaKano, a physician in charge of her health.

What I'm assuming is that she's hospitalized before their school trip as kids because of Uesugi's flashback of A quint and PapaKano. (lack of mother)

While the quints mom is hospitalized, Yots trys to make 1000 paper cranes for her wish to make her mother better, however she doesn't make it in time. Even after her passing, she still continues to make them, maybe out of guilt for not finishing them in time. And now she brings up the "1000 paper cranes" legend because she possibly has another wish she wants to fulfill.

I think Itsuki mentions this not because she lolikano but because she heard the same story we heard when we learned of Uesugi's past. Unlikely that it's Ichika who first found him~!<
