I must be doing something wrong - I have an asus mobo with the screw bracket in place but I don’t have any screws that will screw down into it to hold the drive in place.

I have heard Fastenal recommended. Another solution that I've seen online, but can't verify will work for you, is this insanely overpriced kit from Amazon.

So the threads are broken off in the standoff? Is the standoff removable? Amazon sells replacement standoffs, so you would just need to disassemble (unless you can access the retention screw for the standoff from behind the motherboard), remove the retention screw and the stand off to replace with new, and don't crank down on standoff screws. A half-full turn past finger tight should be plenty to keep pressure on the PCB. Unless your machine is sitting on top of a paint mixer or in a tub of neck massage devices, I can't think of any reason it would back out.

The screws you need should have came with the motherboard. I have the same motherboard and installed an NVMe SSD in the M.2 slot, and cloned to the drive from a 2.5" SSD no problem. After that, I had moved all of my data from 2 HDDs to 2 SSDs.
I currently have an NVMe SSD, and 2 x 1TB SSDs running on the ASUS Z170A
