If you're going to be flying with it, then I highly suggest not using a softshell bag of any kind. Use a hard clamshell style case with two locking points and spinner style combo locks with different combos on each locking point.

Why? Because I used to travel for business around the world and up to 8 months out of every year and airport theft has been a huge problem for me.

I started out with softshell cases, as a young naive business traveler, with no locks and learned my lesson the hard way after a very expensive camera was stolen from the middle of my bag by airport employees on a trip to Dubai.

Then I got a softshell case with interlocking zippers and I put a key lock on the zippers. When I got back from a trip to Las Vegas, my bags were late, ended up on the next flight and an expensive gun scope was missing from my bag. It turns out that airport employees with less than exemplary character carry around keychains with keys for all of the different standard TSA luggage locks.

So I finally said "screw this" and got a rolling, hard clamshell case with a pull handle and built in metal reinforced locking loops on both ends. Then I locked those with rolling combo locks (not spinning). Since then, I've never had anything stolen from a bag again.

As for the interior, there are literally thousands of different solutions. You could get foam that a lot of us tabletop wargamers use and simply make cutouts for all of your deck boxes, playmats, dice bags and water bottles, so they won't get banged around in an airplane cargo hold.

The bag I bought is not in production anymore, but if I were to buy another today for use as you described, then I would buy this one:

But, Chainsaw, that bag is almost $300! Yes, it is. It's probably also going to last longer than you will, and most likely it will be transporting hundreds or thousands of dollars worth of cards and gear that are very important to you, so don't skimp and buy some cheapo camera bag with a zipper. That kind of thing is for going from your house to to game store, not for traveling around the country or the world with your favorite hobby possessions.
