It can sometimes kinda work that way with American comics...

For Deadpool, you can start with the Deadpool Classic books. There are four volumes, with a fifth one coming out soon. It does get a little confusing after that, though. I don't think the Classic books have collected the entirety of the first Deadpool series, which led into Agent X, which I'm not sure has been collected in trade paperbacks either. That then lead into Cable & Deadpool, of which there are three volumes. And then after that you have the current Deadpool series. There are trade paperbacks of those, but the numbering starts at volume 1... which can be really confusing if you want to start at the "real" beginning. Then there's the various mini-series titles which I won't even go into for fear of rage.
TL;DR: Start with the Deadpool Classic books, not plain ol' Deadpool.

Now, with Spider-Man, you have a few options:
You can just get into Ultimate Spider-Man, which is the "updated" or "modernized" version of Spider-Man that started in 2000. Very good stories, and reading them is relatively straightforward as far as Marvel goes. Start at volume 1 and move forward rationally from there for 22 volumes. Then the stupid starts with Ultimate Comics: Spider-Man volume 1, which is basically Ultimate Spider-Man volume 23.
The other options are the Essentials books, which collect the "old school" Spider-Man titles, dating back to the 60s, in big, black-and-white volumes. There are a few titles: Amazing Spider-Man, Spectacular Spider-Man, and Web of Spider-Man which I think will have an Essentials volume coming out later this year. There's also an adjective-less Spider-Man title from 1990 (91?) but I don't know if it's been given the Essentials treatment at all.
Or you could buy this thing and read until you fall over.

Yeah, I believe it becomes Ultimate Comics: Spider-Man, and starts off with a new Volume One. This is the book you're looking for.

Try Spider-Men. It just started, so you can probably find a few #1s, and it's a self-contained story (so-far). They tell you everything you need to know as it comes up. Also, it only goes for 5 issues, so it's a good, short story to start with. You might also want to pick up either the Original Ultimate Spider-Man Volume 1, or you can start with the relaunch with Ultimate Comics Spider-Man. I'd advise you start with the original, since the story line doesn't really start over with the new series, but you should be able to piece together the one or two things you don't get through context and maybe a little Googling. Now, I'm biased towards the Ultimate Universe, so you'll have to get your information regarding the mainstream Spider-Man from someone else.

The story of volume 22 ties in with the Ultimatum event in Ultimatum and Ultimatum:Requiem. (There are mixed opinions on this story, and spiderman isnt the main character so maybe look it up before putting money down)

The story continues on after as Ultimate Comics: Spiderman with Vol. 1 and Vol. 2 and the arc "The Death of Spiderman" in Vol. 3 and Vol. 4 (Personally I think this is probably some of the best 4 volumes of the whole run, up there with the first volume and the first venom volume for me.)

Then Miles' story begins in Ultimate Comics:Spiderman Vol. 2, with 5 collections of that vol.1 vol.2 vol.3 vol.4 and vol.5

That leads to the story in Cataclysm, which is again a main crossover and a spiderman miniseries (all collected in this harcover.

The latest story is the new series, which has only a couple issues out and no trades so far. Its called Miles Morales: Ultimate Spider-man

It has but they changed the name to Ultimate Comics Spider-Man after issue 133 and returned to Ultimate Spider-Man after 20 issues I believe and returned to regular numbering. Here you go.

Ok so, whats the difference between Ultimate Comics Spider-Man vol. 1 and Ultimate Comics Spider-Man vol. 1? I was thinking the second one I linked is the first volume of Ultimate Spider-Man, but that would appear to be Ultimate Spider-Man Volume 1: Powers and Responsibility. So why is there 2 Ultimate Comics #1s?

There's no real lead up (despite the branding), part of what Bendis was trying to do was show how life doesn't set this up. Shit happens. But, you do need to change series. After 22, it gets rebranded and picks up [here] (https://www.amazon.com/dp/0785140999/). Follow the four volumes of that series and you're done. Then you can jump over to Miles' book or stop reading. Bendis does tie up a couple loose ends in Miles' book.

Now, because it was an event, the reason Pete got shot is in a different book, but you still see it happen. You do not need to read that book, and it was kinda shit, but you can if you want to. Though it does appear to be out of print...
